Selection of an Organic Nitrogen Source and Determination of the Parameters for Optimizing the Nutrient Medium for Culturing Streptomyces lavendulae with the Aim of Obtaining Cholesterol Oxidase

Abstract

This work presents a comparative analysis of various sources of organic nitrogen in terms of protein and amino nitrogen content in them for the cultivation of Streptomyces lavendulae in order to produce cholesterol oxidase. The possibility of replacing the traditional component of nutrient media, baking yeast, with alternative sources of organic nitrogen, such as yeast hydrolysate, autolysate, and extract, is being studied. An experiment was conducted to determine the influence of various components of the medium and cultivation conditions on the synthesis efficiency. The negative effect of some yeast substitutes on the enzyme activity has been established. The main parameters for optimizing the nutrient medium and obtaining the maximum amount of cholesterol oxidase have been determined. Among the factors studied, the concentration of the organic nitrogen source, glucose content, cultivation time, and vitamin B supplementation are the most important.
