Nyctanthes arbor-tristis, well known as Parijat or night jasmine, is a revered medicinal plant in Ayurveda, extensively used to manage various aliments including sciatica, arthritis, cough, rheumatism, and parasitic infections. Beyond its aromatic flowers, this plant contains a diverse array of bioactive compounds that contribute to its therapeutic properties. This study aims to characterize the phytochemical composition, physicochemical properties, and evaluate in vitro anti-oxidant and anti-inflammatory potential of its methanolic extract. A methanolic extract of Nyctanthes arbor-tristis flowers was prepared through maceration process and subsequently analysed for phytochemical constituents using chemical tests and thin layer chromatography (TLC). The extract was quantitatively assessed for total phenolic and flavonoid content, followed by comprehensive in vitro evaluation of its antioxidant and anti-inflammatory activities. Phytochemical analysis confirmed the presence of alkaloids, flavonoids, phenols, saponins, and glycosides. Quantitative analysis revealed the content of phenolic (48 μg/ml) and flavonoid (38 μg/ml). The extract exhibited potent antioxidant activity in hydrogen peroxide (IC50 = 38.77 μg/ml) and nitric oxide (IC50 = 46.01 μg/ml) scavenging assays. Furthermore it demonstrated potent anti-inflammatory effects in the egg albumin denaturation assay (IC50 = 30.08 μg/ml) indicating significant protein stabilization capacity. The study provides compelling evidence for Nyctanthes arbor-tristis flowers as a therapeutically valuable plant resource. The high phenolic and flavonoid content correlates with strong anti-oxidant and anti-inflammatory activities, suggesting promising applications in developing natural therapeutics for oxidative stress-related inflammatory disorders.
